چکیده

China isolates of Phytophthora parasitica var. Nicotianae have been divided into three large pathogenic groups (PGS) (Hv(A), Iv(B), Wv(C)) which came from Yunnan, Shandong and Liaoning. These three districts belong to main tobacco regions in China: 15 isolates derived from Yunnan of the southwest, 13 isolates from Shandong of the east and 20 isolates from Liaoning of the northeast. PGS were determined through pathogenic assays in greenhouse studies with three tobacco cultivars. The isolates were analyzed by means of the random amplified polymorphic DNA (RAPD) method. All tested isolates could be differentiated into four cluster groups. Each cluster group contained different PGS did not belong to the same cluster group. The conclusion shown that there was no significant different in RAPD pattern among highly virulence, intermediate virulence and weakly virulence groups in China. Therefore, the similarities and difference in handing patterns by RAPD could not be a useful molecular tool in identification and phytogenetic studies of the PGS of the same pathogen from the difference tobacco districts.

Random amplified polymorphic DNA (RAPD) fingerprinting of Mycoplasma gallisepticum isolates from chickens

Ten field isolates of Mycoplasma gallisepticum (MG) from different geographical areas of Iran were analyzed by random amplification of polymorphic DNA (RAPD). RAPD analysis produced reproducible banding patterns on the basis of which various distinct amplification patterns could be detected. MG isolates compared with reference strains (S6 and Mg SS) and vaccine strain (ts-11) and demonstrated...

Characterization of Pseudomonas aeruginosa in Burn Patients Using PCR- Restriction Frag-ment Length Polymorphism and Random Amplified Polymorphic DNA Analysis

One of the major opportunistic pathogens in patients with burn injuries is Pseudomonas aeruginosa, which causes severe infections in burned patients. The objective of the study was to examine the molecular epidemiology of P. aeruginosa colonization in the burn unit of Shahid Motahari Hospital in Tehran, Iran. Restriction fragment length polymorphism (RFLP) and random amplified polymorphic DNA (...

IDENTIFICATION OF LEISHMANIA SPECIES FROM DIFFERENT PARTS OF IRAN USING A RANDOM AMPLIFIED POLYMORPHIC DNA IN HUMANS, ANIMAL RESERVOIRS AND VECTORS

In this study, we used Random Amplified Polymorphic DNA (RAPD) for identification of 17 isolates of Leishmania from the skin and reticuloendothelial system of humans, animal reservoirs (rodent and dog) and sandflies in various parts of Iran in the last decade. Fifteen species have been confirmed by isoenzyme characterization by the London School of Hygiene and Tropical Medicine and Shiraz ...

PCR amplification of species-specific DNA sequences can distinguish among Phytophthora species.

We used PCR to differentiate species in the genus Phytophthora, which contains a group of devastating plant pathogenic fungi. We focused on Phytophthora parasitica, a species that can infect solanaceous plants such as tomato, and on Phytophthora citrophthora, which is primarily a citrus pathogen. Oligonucleotide primers were derived from sequences of a 1,300-bp P. parasitica-specific DNA segmen...

Analysis of Phytophthora parasitica by retrotransposon-derived DNA fingerprinting

Phytophthora parasitica, being able to attack a wide variety of plants, is a very important plant pathogen in Taiwan. It has been shown that isolates of P. parasitica from tobacco, dieffenbachia, and loquat differed significantly from other isolates in morphology and pathogenicity and were recognized as “atypical” types of the fungus. In the current study, a partial sequence of the reverse tran...
